Clenbuterol is a direct-acting selective beta2 receptor agonist. Each tablet contains 40 microgram of micronized Clenbuterol Hydrochloride, according to Ph.Eur.5.5. Each tablet also contains following excipients: lactose monohydrate, sodium starch glycolate, polyvidone 25,000, microcrystalline cellulose and magnesium stearate. The 40 mcg tablet do not contains any colouring agent. Clenbuterol differs from other β2-specific sympathomimetic by the low effective dose, long biological half-life, rapid and complete absorption by oral administration. It is used as a bronchodilator in the management of reversible airways obstruction such as asthma and certain cases of chronic obstructive pulmonary disease. Athletes and bodybuilders use the drug due to its thermogenic and anti-catabolic effects. Core temperature increase of 0.5oC yields around 5% increase in calories expenditure.
Clenbuterol binds preferentially to adrenoreceptors on cell membranes of the bronchi, which causes activation of adenylate cyclase in smooth muscle cells, leading to bronchodilation. The substance inhibits histamine release from the mast cells in the lungs, and enhances mucociliary clearance. After oral administration, clenbuterol is readily absorbed and maximum plasma concentrations reached within 2 hours. The substance is rapidly distributed in tissues and metabolised primarily by the liver.
Approximately 45% of the dose is eliminated as Clenbuterol through urinary tract.
The kidneys excrete 70 – 91% of the total dose, and the remainder is eliminated in the faeces (6 – 15%)
Dosage and administration
Usual daily dose of clenbuterol is 40 mcg. The dose should increase progressively during the whole week of the course. It is recommended to take clenbuterol in the morning, because it can cause sleeplessness, if taken before sleep. Moreover, the efficacy of the drug is higher in the first half of the day, because of human circadian cycle. Indications:
For prolonged treatment of bronchospastic syndrome in mild and moderate atopic and non-atopic bronchial asthma; strain-induced bronchospasm; in the complex therapy of chronic spastic bronchitis and COPD.
Beta-blockers antagonize clenbuterol effects. In concomitant administration with antidiabetics, their dose should be increased due to stimulation of glycogenolysis by clenbuterol. Co-administration with cardiac glycosides, MAO-inhibitors and theophylline may cause heart rhythm problems.
The effect of clenbuterol is potentiated by tricyclic antidepressants, some antihistamines and levothyroxine. The effect is reduced by some antihypertensive products (guanethidine). Halothane and other halogenated hydrocarbon anesthetics and cyclopropane sensitize myocardium and may potentiate the arrhythmogenic effect of в 2-sympathomimetics.
The use of Clenbuterol Hydrochloride can be followed by possible side effects that can be severe. Such effects are most commonly associated with high dose abuse and far beyond recommended periods of use. The severe side effects of clenbuterol include high blood pressure, irregular heartbeat, trembling and even panic. Some studies have shown that Clenbuterol abuse can also lead to cardiac hypertrophy, which could potentially lead to death.
